Growth inhibition of human tumor cell lines of different origins with medicinal mushroom preparations

For centuries medicinal mushrooms have been widely used in traditional medicines for hepatitis, cardiovascular illnesses, cancer, and diabetes treatment ; however, in modern medicine and pharmacology, the mechanisms of the biological effects of mushroom preparations have not been systematically investigated. Biological effects of compounds present in mushrooms are multidimensional, and their mechanism of action, once they reach the cell, includes a cascade of events such as the inhibition or induction of expression of certain proteins as well as the modification of basic metabolic processes in the cell and differentiation of the redox status of the cell. In this work, cytotoxic, anti-oxidative and pro-oxidative nature of blended mushroom extracts (Lentifom, Superpolyporin, Agarikon, Agarikon+, Agarikon.1 and Mycoprotect.1) and extracts of several single mushroom species (Boletus edulis, B. regius, Scutiger pes-caprae, Russula virescens, Cortinarius violaceus, Cornularia cornucopiae, and Pleurotus tuberregium) were examined on model test systems including cell lines isolated from human ductal breast tumor, small cell lung cancer, non-small cell lung cancer, and colon cancer. Obtained results will reveal if this extracts have different cytotoxic effects on tumor cells originated from different tissues, and if so, which cell line is the most susceptible to growth inhibition. These investigations will provide insight into the antitumor effects of mushrooms extract preparations and their overall influence on different biological test systems. In the first phase of investigation, cytotoxic effects of different concentrations of mushroom extracts will be determined. Neutral red method will be used for the detection in the possible destabilization of the membranes in the cells, and the MTT test will be used for the determination of changes in mitochondrial metabolism. Concentrations and conditions under which mushroom extracts exert their anti-oxidative and pro-oxidative nature will be determined on different cancer cell lines isolated from the lung, breast, and colon cancers. From this data, it will be possible to estimate what is the connection between maximal anti-oxidative/pro-oxidative effects of mushroom preparations and time/concentration of exposure.
