Fluorescent chromosomal banding in Picea omorika and P. abies
Cytological investigation were conducted on Picea omorika, an endemic species to the Balkan region. The chromosome number was confirmed to be 2n=2x=24, and the chromosomes were metacentric, except two pairs with submetacentric centromeres. The chromosomes were differentially stained by chromomycin A3 (CMA) and Hoechst 33258. Well distinguished bands were obtained by CMA, while bands visible through Hoechst staining were negative with slightly shining telomeric regions. Picea omorika CMA-banding patterns were compared with banding patterns of related species, Picea abies. Sixteen CMA-bands were distinguished in the P. omorika karyotype, in contrast to 14 CMA-bands appearing on P. abies chromosomes. The majority of CMA-bands in both Picea species were interstitially located with only a few bands located proximal to the centromere. In addition, interstitial CMA-bands appeared as two thick bands at both adjacent sites of secondary constriction connected by CMA-positive thin thread in both species.
