Cytogenetic analysis of black pine transgenic roots
Cytogenetic analyses were conducted on seven-day-old adventitious roots from on European black pine (Pinus nigra) six-veek-old epicotyl explants infected with Agrobacterium rhizogenes, strain 8196 or A. tumefaciens, strain C58C1 harboring the Ri plasmid (pAr4b). The roots formed in an axenic culture on liquid 1/2xGD medium supplemented with 1% sucrose and 500 mg/l carbenicillin were not phenotypically different from the untransformed seedling roots. Mitotic and chromosomal abnormalities, like anaphase and telophase bridges, lagging chromosomes, fragments and micronuclei were observed in all roots, but in a low frequency. There were no changes in chromosome number. The frequency of total aberrations was not significantly different when compared with analyses of noninfected seedlings. No changes were found in the heterochromatic region stained with chromomycin A3 between transformed and normal roots.
