Expression of bone morphogenetic proteins during inflammatory bowel disease
Bone morphogenetic proteins (BMPs) are a large group of structurally related proteins that belong to the transforming growth factor- (TGF-) superfamily. Along with their osteoinductive function they play an important role in the development and healing of different non-skeletal tissues and organs. Alterations in BMPs expression and BMP signaling pathway have been associated with the pathological conditions linked to several human diseases such as inflammatory bowel disease (IBD) and cancers. To investigate the potential involvement of BMPs in inflammatory bowel disease this study examined the expression of BMPs and their signal molecules (Smad proteins) through RT-PCR and immunohistochemical analysis in a rat inflammatory bowel disease model. This study was designed to explore the expression of BMP-2, -4, -6, -7 and their signaling molecules Smad1, Smad5 and Smad8 in rat colon during acute and chronic phases of inflammatory bowel disease as well as after BMP-7 treatment. IBD was induced by intracolonical administration of trinitrobenzenesulfonic acid (TNBS). After the IBD induction, the rats were treated with BMP-7 (100 g/ml) and sacrificed on the 2nd, 5th, 14th, and 30th day after the TNBS induction. During IBD the expression of BMPs was present in colon samples with a slight incensement of BMP-2 and BMP-7 expression in the chronic phase of the disease. BMP-7 treatment moderately enhanced expression of BMP-4, -6 and -7 and suppressed the BMP-2 expression. Also, we found increased expression of Smad proteins for intracellular BMP signaling (Smad1, Smad5, and Smad8) during IBD as well as after BMP-7 treatment. The expression pattern of BMPs and Smad proteins during IBD as well as after BMP-7 treatment suggests an important role of BMPs and BMP signaling pathway in the control of tissue damage and healing during disease.
