Storage product accumulation during the maturation of Picea omorika (Panč.) Purk. somatic embryos

Different developmental stages of somatic embyos of the established embryogenic culture, and mature zygotic embryos of Picea omorika were used for light and electron microscopy. The content of storage products (starch, proteins) was quantified by image analysis and SDS-PAGE electrophoresis. By using image analysis and microscopic in!vestigation we monitored the accumulation of storage reserves during the maturation of Picea omorika somatic embryos. The early stages of embryo development were characterized by a high starch content and the initiation of storage protein accumulation. In mature somatic embryos the starch content decreased, while protein and lipid content continuously increased. The presence of exogenous ABA in the culture medium caused a higher storage protein content in the mature somatic embryos, compared to mature zygotic embryos. The results obtained by using image analysis correlated to the results of SDS-PAGE analysis of proteins.
