Antioxidant and anticholinesterase evaluation of water extract and essential oil from milefollium (Achillea Millefolium L.)

The aim of the study was to investigate antioxidative and inhibitory properties on enzyme acetylcholinesterase (AchE) by millefolium (Achillea Millefolium L.) water extract and its essential oil. The chemical composition of the essential oil was identified using coupled system gas chromatography-mass spectrometry (GC-MS). The The major components identified in the oil are bycyclic sesquiterpenes kamazulene and β kariofilene. Antioxidative activities of the plant water extract and its essential oil were analyzed using the 1, 1-diphenyl-2-picrylhydrazyl radical scavenging method (DPPH) and Briggs-Rauscher (BR) reaction method. The plant water extract and its essential oil were also tested for AChE inhibitory activity using Ellman’s colorimetric method. The results obtained using DPPH method showed that the plant water extract has better antioxidative propperies in comparison to its essential oil. The measurement of the essential oil antioxidative activity based on BR reaction showed that this method is not suitable for lipophilic antioxidant. However, the results for the water extract showed that antioxidative activities are linearly dependent on antioxidant concentration. The maximum inhibition against AChE for essential oil was 27, 4% at concentration of 0, 8 mg/mL, and for the water extract 50% inhibition was reached at concentration of 55, 78 mg/mL.
