Synthesis and anti-inflammatory activity of 1-thia-dibenzo[e, h]azulenes modified at position 8
1-Thia-dibenzo[e, h]azulene derivatives were found to be new potent molecules with antiinflammatory activity. One of the main research goals was to investigate the way modifications of different sites at 1-thia-dibenzo[e, h]azulene scaffold affect biological activity. The most obvious and synthetically very interesting modification appeared to be exchange of different heteroatoms (O, S and N) and CH2 group at position 8. We have searched for the most efficient and economic route, which would use the common synthetic intermediate as early as possible. Antiinflammatory activity of target molecules was determined in vitro and in vivo as inhibition of the key proinflammatory cytokine tumor necrosis factor alpha (TNF-alpha).
