Preliminary optimization of the binding and elution of viroid single-stranded circular RNA on monolithic chromatographic supports
Single-stranded (ss) (1, 2) and double-stranded (ds) RNA molecules (2, 3) have been previously purified by monolithic chromatographic supports. Here we present preliminary optimization of binding and elution of viroid circular ssRNA represented by Potato spindle tuber viroid (PSTVd). This type member of family Pospiviroide is a pathogenic ssRNA molecule that contains 359 nt and forms rod-like tertiary structure. The viroid RNA was monitored and quantified in the elution fractions by a PSTVd specific real time RT-qPCR assay. Final purpose is separating pure viroid from host plant RNA which was monitored by a cytochrome oxidase (COX) specific RT-qPCR assay
