Temperature impact on uptake and survival of Listeria monocytogenes within Acanthamoeba castellanii
Listeria monocytogenes is widespread in nature and has a possibility to survive in different hursh conditions, but the way of its environmental transmission and ecology is still uncelar. It is known that free-living amoebae may act as a predators, as well as an environmental reservoirs and vehicles for the transmission of different facultative and obligate intracellular pathogens. In this study, the interaction between the protozoan Acanthamoeba castellanii (ATCC 30234) and Listeria monocytogenes (EGD strain) was studied by viable counts and gentamicin assay, under different temperature regiment. At 22 °C listeria was internalized into A. castellanii but after 4h of co-cultivation we were not able to recover any viable intracellular bacteria. In co-culture incubated at 35 °C, listeria was ingested but was completely unable to establish an intracellular lifestyle within amoeba trophozoites. At the same time extracellular counts of listeria were about 1000-fold greater in the presence of amoebae compared with that in their absence. In conclusion, co-culture experiments under different laboratory temperatures suggest that A. castellanii act as a predator on internalized listeria and that different temperatures have minor effect on its intracellular survival. However, the presence of amoeba positively influenced extracellular bacterial growth and multiplication. Finally, the data presented in this study suggest that free-living amoebae, such as A. castellani, probably do not serve as a natural reservoir for L. monocytogenes.
