Genetic cartography of Oak populations in Croatia based on Cp DNA markers
Oaks from Quercus sensu stricto group were analysed. Four chloroplast DNA fragments were amplified by PCR. These fragments are digested with restriction enzymes, two fragments with HinfI and two with TaqI. Fragments obtained by digestion were separated by electrophoresis on polyacril-amide gels. Results were scored based on the position of bands on gels. Results for Croatian populations are interpreted in comparison with data obtained by other researchers within Fair Oak project. Cp DNA haplotypes were strongly geographically structured and thereafter enable us to reconstruct its colonization routes in Croatia. Upon glaciation Croatia was colonized from three directions. Slavonian oaks are of eastern origin, probably from a Balkan refugium. Oaks of Dalmatia, Lika and central Croatia were spread from Apenninic refugium across today's north Adriatic. Finally, oaks from western Croatia are of yet not fully clear origin. They may come from a new identified refugium on west Croatian coast or from east Pyreneans. In this investigation some clearly introduced populations were also detected, based on izs haplotypes not found in neighboring populations.
