Analysis of three mouse genes identified by gene trap
In order to identify mouse genes involved in embryo develpment we applied the gene trap method. Embryonic stem (ES) cells were genetically modified by a nonhomologous construct containing the splice acceptor and fused promoterless genes lacZ and neoR. Both inserted genes can be active if construct integrates within the endogenous gene, trapped in this way. This enables to identify trapped gene, to characterise its expression pattern and to get insight in the gene"s function as only part of the gene 5" from the construct will be active. We analyse three mouse lines with the gene trap integration within three different mouse genes. 5"RACE showed that two of the genes are novel mouse genes. Both genes are expressed from 9.5-day embryos onward in the heart and the neural tube. The first gene named kosenic is expressed only in the developing trabecular system of the heart and in the column of cells located in he ventral part of the neural tube. The expression of the second gene named lobel is similarly restricted to the cells within the ventral part of the neural tube, but it is present in the whole heart. Both genes are expressed in the adult brain. The third trapped gene is Nol1, coding for the proliferation associated nucleolar protein p120. Within the cell its expression is very strong in the nucleolus. It is expressed in all structures of the developing embryo, but not in all cells. Th intercrosses between heterozigous animals resulted with no living homozygous offspring.
