Growth morphogenesis and genome instability of Vicia faba L. cultures
Callus cultures were obtained from explants of immature (strains VFS1 and VFZ6) embryos of Vicia faba L. on various media. The effects of some hormonal and environmental factors on growth were determined by fresh weight values. The maintained callus cultures were grown most satisfactory either on RM-1962 medium with 3% sucrose, 0.92 µM 2, 4-D and 1.0 µM kinetin, or on B5 modified medium with 3% sucrose, 2.3 µM 2, 4-D and 25.0 µM kinetin. On the last medium the cultures grew more uniformly, being yellowish and without necrosis. The optimal illuminatio intensity was about 500 lx. After being cultured for nearly 3 years, 8-33% of VFS1 callus cultures were able to regenerate roots, when transfered either to RM-1962 half strength with 5.37 µM NAA, or on media without 2, 4-D, as well as on media with only kinetin (in various concentrations). At the same time the dependence of genome instability on various nutrient media as well as on the regimen of subculturin was studied. Karyological analyses of various strains, in early as well in late subcultures, have shown that all strains contain populations of cells with various ploidy and aneuploidy levels. A high percentage of polyploid cells was observed when the tissue was longer in culture without being subcultured.
