Consistency of instability in chromosome complement of long-term Vicia faba cultures
Vicia faba callus line, initiated from explants of immature embryos and maintained on appropriate media supplemented with auxin (2, 4-D) and kinetin, has been cytologically examined from 9th to 20th subculture (age 16 to 20 months). Using Giemsa C-banding procedures numerical and structural changes were noticed. Frequency of polyploidy decreased with the age of culture ranging in value from 12x to 2x corresponding aneuploids (hyperdiploids or hypotriploids). In the 9th subculture all investigated metaphases had 10 (±1/+1 extra chromosome/triploids or tri-aneuploids), while in the 15th subculture the observed karyotypes contained 14 chromosomes (hypotriploids). All karyotypes of both the 9th and the 15th subculture indicate, however, that the consistency of instability of chromosome number and structure depends on the type of chromosomes.
