IDENTIFICATION OF WORLD WAR II MASS GRAVE VICTIM BY RARE Y CHROMOSOM STR LOCI MUTATIONS

We present the process of identification of skeletal remains from a mass grave found in 2009. on Daksa, small island near Dubrovnik, Southern Croatia. The 50 skeletions remains were thought to be those of civilians killed in the Second World War, among them were group of a few friars. Excavation of site was done according to archeological procedures. DNA was isolated from bones and teeth samples using standard phenol/chloroform/isoamyl alcohol extraction. DNA quantification and amplification were done according to the recommended protocols. DNA typing was performed using Y-chromosome STR analysis. These results were matched with those of DNA analysis of blood samples collected from the relatives of supposed missing persons. Cross-matching, using Y-STR systems, gave positive identifications. In this report we present the identification of one skeletal remain by rare Y chromosome STR loci mutations. DNA male profiles were obtained from evidence bone/teeth samples and compared with paternal lineage relatives' blood samples using Y-STRs. We found a father/son pair with double peaks at DYS437 locus as well as with null allele at DYS448 locus observed in the same father/son pair. While observing a null Y-STR locus should not represent a problem for profile interpretation, more than one peak at one or more loci could lead up to a deep misinterpretation of a profile and from which kind of sample the profile come out. The same rare mutations and their interpretation have been already reported. Multiple variations (deletion or duplications), even if rare, can happen in the same single-source sample and these examples should be a reminder before drawing premature conclusions. This should focus the attention on forensic interpretation of Y-haplotype profiles, because multiple alleles at various loci do not forcibly indicate that the sample originates from a mixture. This also showed the importance of DNA identification methods and their application in case work.
