Korelacija aktivnosti glutation S-transferaze plijesni Aspergillus flavus s opsegom sinteze aflatoksina
U istraživanju je ispitivana aktivnost enzima glutation S-transferaze (GST) u odnosu na sintezu aflatoksina plijesni Aspergillus flavus, soj NRRL 3251 na različitim hranjivim podlogama. Korištene su GMS i PMS tekuće podloge koje sadrže različit izvor ugljika. Utvrđena je znatno veća količina aflatoksina na glukozom bogatoj GMS podlozi. Na PMS-u je izmjereno maksimalno 23, 09 ppb ukupnih aflatoksina, dok na GMS-u maksimalan udio istih iznosi 362, 14 ppb. Za analizu aflatoksina korištena je standardna metoda – imunoafinitetno pročišćavanje u koloni s HPLC analizom. Udio proteina veći je na PMS podlozi zbog prisutnosti peptona. Prema dobivenim rezultatima istraživanja, nije moguće utvrditi na kojoj je hranjivoj podlozi aktivnost GST-a veća. Također, nije utvrđena statistički značajna korelacija između sinteze aflatoksina i aktivnosti GST-a.

Correlation between Glutathione S-transferase activity and aflatoxin synthesis in Aspergillus flavus
Correlation between glutathione S-transferase activity (GST) and aflatoxin production by the mould Aspergillus flavus, strain NRRL 3251, grown on different culture media, was examined. GMS and PMS liquid culture media, containing diverse carbon sources, were used. A greater amount of total aflatoxins was determined in glucose rich GMS liquid Maximum total aflatoxin concentration in PMS medium was 23.09 ppb, while 362.14 ppb was measured in GMS. Standard methods of aflatoxin clean-up and detection were applied – immunoaffinity column chromatography combined with HPLC analysis. The quantity of protein was higher in PMS medium due to peptone content. According to the obtained results, it was not possible to estimate which culture media supported higher GST activity. Additionally, no statistically significant relationship between GST activity and aflatoxin production was determined.
