Macrolide anti-inflammatory activity in lipopolysaccharide (LPS) induced inflammation models
Macrolides, apart from their antibacterial activity, suppress proinflammatory cytokine production and migration of inflammatory cells. Therefore we tested antiinflammatory activity of erythromycin (ERY), clarithromycin (CLA) and azithromycin (AZI) on LPS induced plasma tumor necrosis factor alpha (TNFa) production and lung neutrophilia. CLA significantly reduced TNFa concentration in plasma as well as total cell number, percentage of neutrophils and TNFa concentration in bronchoalveolar lavage fluid (BALF) and inflammatory cell infiltration into lung tissue. ERY exhibited an effect similar to CLA, while AZI markedly, although not significantly, decreased total cell number in BALF. Our results suggest that macrolides could differ in their acute antiinflammatory activity.
