INFLUENCE OF VACCINE AND WILD-TYPE MUMPS VIRUS INFECTION ON TYPE I IFN INDUCTION CASCADE
Background. We investigated mumps virus (MuV) strain differences following infection of distinct human cell types chosen as the representative targets during natural infection. Intent was to examine the ability of MuV strains (vaccine and wild-type) to trigger an innate immune response in these cells by comparing induction of viral RNA sensors and selected components in type I IFN induction cascade. Methods. The cell monolayers (A549, SHSY-5Y, MRC5) were infected with different MuV strains. The total RNA was extracted, reverse transcribed and the relative expression of mRNA for IFN-activation associated genes was measured by qPCR. Results. Irrespective of the cell lines used in our experiments, we show a similar trend of expression levels for analysed genes. Generally lower levels of mRNA expression were obtained upon stimulation of infected cells with poly(I:C) compared to poly(I:C)-stimulated uninfected cells, especially for wild-type MuV strains. Significant difference in gene induction was observed between the group of vaccine and wild- type MuV strains, latter inducing genes to a far lesser extent. We further investigated defective interfering particles in vaccine and wild-type MuV strain and their contribution to the induction of cellular response. Our preliminary results indicate that influence of these particles depends on a cell type chosen for infection. Conclusions. The results of our study demonstrate that MuV vaccine strains are stronger inducers of IFN-activation associated genes than wild-type MuV strains.
