Ammodytoxin content screening in the Vipera ammodytes ammodytes venom samples
Snake venoms are complex mixtures of more then 100 proteins, peptides and other compounds which biochemical and pharmacological variability has been demonstrated. These mixtures have been used as antigens for immunization of antivenom-producing animals (horses or sheep). Ammodytoxins (Atx) are potent neurotoxins, and the most toxic compounds isolated so far from the Vipera ammodytes ammodytes (Vaa) venom. The content of Atx in the Vaa venom is a good measure of its immunogenicity and toxicity. The development of the cation exchange chromatographic method using CM-CIM for extraction and fast separation of highly basic venom components (including Atxs) will be presented. The usefulness of the method for the screening of venom samples, in comparison to several other methods, including capillary gel electrophoresis-on-a-chip and newly established ELISA, will be reported and discussed.
