Population genetics of 17 Y-short tandem repeat loci in Southern Croatia

The increased use of human Y-short tandem repeat (STR) markers in forensic analysis has resulted in a collaborative effort to collect haplotype data from different populations and to create a reference database of European haplotype data. The aim of this study was to perform a Y-STR haplotyping on a population sample of Southern Croatia. We carried out a statistical analysis of the data from previously performed genetic analysis collected during the routine forensic work by the Forensic Science Centre “Ivan Vučetić”. A total of 220 unrelated healthy men from Southern Croatia were selected for the purpose of this study. Genomic DNA was extracted using Chelex procedure from FTA cards. Y-chromosomal STRs were determined using the AmpFlSTR Yfiler PCR amplification kit. The haplotype frequencies were determined by direct counting and analyzed using Arlequin 3.1 and AMOVA analysis calculated with YHRD online analysis tool. The overall locus diversity for entire sample was 0.554, and varied from 0.203 for DYS392 to 0.765 for DYS458. A total of 197 distinctive haplotypes were recorded, while 181 were registered only once, producing a total discrimination capacity of 82.3%. The AMOVA analysis suggested the most resemblance of the Southern Croatian population to other Croatian samples, while the closest neighboring population was that from Mostar, Bosnia and Herzegovina. The results also suggested large differences from the other neighboring countries, suggesting a distinctive genetic background of the investigated population. Further attempts for a thorough genetic characterization of Croatian population are under way.
