Genetic purity testing of maize seed using SSR markers
Two experiments were carried out in order to study the possibility of application of SSR markers in maize seed production. In the first experiment phenotypically typical, atypical and suspicious plants of two hybrids and their parental components were taken from the field and analysed for three SSR loci (umc1545, umc1887 and phi050). All phenotypically typical plants possessed typical genotype, except one male plant. All phenotypically suspicious plants possessed typical genotype, except in two cases. In the second experiment twelve plant tissue bulks were created with different percent ratio between true-to-type inbred line and an inbred line, which served as impurity, in order to simulate unwanted crosspollination and mechanical impurities. The objective of this experiment was to study how appropriate are DNA samples obtained from tissue bulks in detecting the contamination of true-to-type DNA with foreign DNA. The percent of impurity varied from 4 to 30 %. The bulks were analysed at two SSR loci (umc1304, phi037). Minimal detectable percent of impurity in this study was 7%, represented by a tissue bulk containing one atypical plant (foreign DNA) in the bulk comprised of 15 individuals.
