How reliable is LABTypeSSO B Locus for high resolution B*27 typing?

The HLA-B27 is one of the most extensively studied HLA genes because of its remarkably strong association with ankylosing spondylitis and other spondyloarthropaties. The most commonly used methods for the determination of HLA-B27 are CDC, flow cytometry and PCR-SSP. Recently developed bead array method based on SSOs bound to fluorescently coded microspheres has been designed for low resolution typing. Single nucleotide changes can be detected in some cases. The aim of this study was to compare bead array method (LABType SSO B Locus, One Lambda) with high resolution SSP (HLA-B*27, Olerup SSP) in order to validate B*27 allele level typing using LABType SSO B Locus. Additionally, we wanted to determine the frequency of rare B27 alleles in studied groups due to the recent discovery of a rare HLA B*2730 allele in Slovenian population. This allele was subsequently also found among Croatians. Both methods were used for typing a total of 116 individuals (69 Croatians and 47 Slovenians). LABType SSO typing revealed HLA-B*27 typing at high resolution typing level for 20 samples as B*2702 and one for each of B*2707, B*2709, B*2730. Medium resolution typing was obtained for 45 individuals as B*2702/30 and for 48 samples as B*2703/05/13/17/19. All high resolution LABType SSO results were confirmed by high resolution HLA-B*27 PCR-SSP method. All B*2702/30 individuals were retyped as B*2702 while all B*2703/05/13/17/19 were determined to be B*2705. LABType®SSO B Locus proved to be reliable for medium or high resolution B*27 typing in groups of alleles studied. The only individual carrying B*2730 was Croatian (0.74%) which may suggest that the allele originates closer to Croatia than to Slovenia.
