Serum platelet-activating factor acetylhydrolase activity and levels of CRP and At-oLDL in uremic patients on hemodialysis (CROSBI ID 561731)

Serum platelet-activating factor acetylhydrolase activity and levels of CRP and At-oLDL in uremic patients on hemodialysis
Patients on hemodialysis (HD) and peritoneal dialysis (PD) are at risk for premature atherosclerosis. The aim of the present study was to evaluate whether platelet-activating factor acethylhydrolase (PAF-AH) and paraoxonase (PON) as well as PON phenotyping were altered in patients on HD and PD and the relationship of enzyme activities with CRP and At-oLDL in comparison with control group. We studied 139 patients on HD and 38 patients on PD in comparison with 234 sera of a control group (CG).PAF-AH activity was measured with 1-myristoyl-2-(4-nitrophenylsuccinyl) phosphatidylcholine and PON activity with paraoxon. Sera of the patients and control subjects were assigned to PON phenotypes based on the basal PON activity distribution. In comparison to the CG (median 326 U/L), higher PAF-AH activity were in HD (median 352 U/L, p=0.012), and PD (median 368 U/L, p=0.024), and lower PON activity were determined in HD (median 58 U/L, p<0.001), and PD (median 77 U/L, p<0.008). A higher frequency of AA paraoxonase phenotype was detected in patients on HD (CG 60% vs. HD 77%, p=0001). Positive correlation were between the At-oLDL level and CRP concentration (r= 0.2250, p= 0.0078) and negative correlation between PON and At-oLDL concentration (r=-0.2150, p=-0.1880) in patients on HD. The change of PAF-AH and CRP were more pronounced in patients assigned to AA paraoxonase phenotype group. Positive correlation between At-oLDL and CRP and negative correlation between the enzyme activities and CRP in patients on HD confirm the role of inflammation in atherosclerotic process.
