Cryopreserved human monocytes for the in vitro test of monocyte spreading inhibition
We used human peripheral blood monocytes as indicator cells for the in vitro test of cellular immunity. We tried simple cryopreservation method without gradual cooling and storage at -198 ºC and placed the cells directly at -80 ºC. We first determined the functionally test of monocyte spreading. Thawed monocytes spread less well than fresh cells, but after 12 weeks the percentage of spread cells fell below 50%. In evaluation of immunosuppressive potential of the horse-anti.human lymphocyte globulin the monocyte sporeading indices were somewhar lower after preservation, most likely due the lower spreading ability of the thawed monocytes. The cryopreserved cells were not usable in the test of monocyte spreading inhibition after preservation for more than threee weeks. They were in clumpd and the number of cells in the suspension decreased, so that it made their use impractical.
