Lymphocyte phenotyping in BAL. How to avoid pitfalls, inherent in using the conventional lymphocyte gate?
Bronchoalveolar lavage (BAL) obtained fluiid contains cells recovered from the lumen of the lower respiratory tract. Due to the heterogeneity of BAL cells, a potential problem in analysis by flow cytometry lies in a proper gating of selected population. The inappropriate gate may lied to exclusion of cells of interest and inclusion of unwanted cells. A 38-year old man with a diagnosis of chronic pulmonary sarcoidosis was followed-up in the period from August 1990 through May 1995 and tested 11 times. Lymphocytes in BAL were characterized by a panel of monoclonal antibodies. Double color immunofluorescence analysis was carried out using a FACScan after establishing a lymphocyte gate. The principles and options as well as the advantages and disdvantages of various methods in selecting lymphocyte gate will be discussed.
