Laser Capture Microdissection and PCR for analysis of HPV Infection

Human papillomavirus (HPV) infection is considered as one of main factors involved in the pathogenesis of squamous cell carcinoma (SCC) and adenocarcinoma (ADC). Our previous study revealed a high prevalence of high-risk (hr) HPV-DNA in examined cervical lesioons (>90%). HPV-16 predominated in SCC (65%) and HPV-18 in ADC (67.6%). The prevalence of particular hr-HPV types in adenosquamous carcinoma (ADSQ) is controversal as well as the presence of different HPV types within this tumor. Tha aim of this study was to detect hr-HPV DNA and to determine the HPV type in microdissected tissue of 4 cases of cervical adenosquamous carcinoma using lase capture microdissection (P.A.L.M.) followed by DNA extraction and qualitative PCR. Our results confirm the associtaion of certain hr-HPV types to different histological entity of ADSQ and importance of microdissection in molecular diagnostics.
