Insulin administration in the mild hyperglycaemia changes expression of proinflammatory adhesion molecules on human aortic endothelial cells

The induction of adhesion molecules on the surface of endothelial cells is one of the first steps in a high glucose-mediated endothelial dysfunction in diabetic patients. Influence of insulin administration in condition of elevated glucose concentration on the E-selectin, vascular adhesion molecule-1 (VCAM-1), and intracellular adhesion molecule-1 (ICAM-1) expression on human aortic endothelial cells (HAEC) was investigated. HAEC were cultured 4 h in a Medium 200 with low serum grow supplement, homocysteine (7 μM) and glucose in range of concentration (5.5, 8.0, 12.0 and 16.5 mM). Expression of CAMs were analysed by flow-cytometry using monoclonal antibodies. Controls were CAMs expression in the medium with a physiological glucose concentration (5.5 mM). The influence of insulin (1 mUI/mL) was analyzed in combination with each glucose concentration. Incubation of HAEC with 8 mM glucose increased expression of E-selectin for 2-fold, ICAM-1 for 47% and VCAM-1 for 3-fold. Expression of E-selectin was 23% lesser, ICAM-1 was 64% higher and VCAM-1 was 41% when cells were grown with 12 mM glucose. Incubation of HAEC with 16.5 mM glucose decreased expression of E-selectin for 88%, ICAM-1 for 12% and VCAM-1 for 43%. Addition of insulin was not changed expression of CAMs expression in a medium with 5.5 mM glucose. In conditions of elevated glucose concentration (12 mM), addition of insulin resulted in significantly decreased expression of E-selectin (27%) and increased VCAM-1 (23%) expression. In conclusion, moderate higher glucose concentration increased E-selectin, ICAM-1 and VCAM-1 expression in HAEC. Insulin administration in the mild hyperglycaemia condition reduces the expression of the proinflammatory adhesion molecule E-selectin which could contribute in slowing of macrovascular complications development in diabetic patients.
