Lectin receptors and regulatory peptides in bovine abomasum
The binding properties of lectins Con A, PNA, WGA, DEA and UEA I were examined histochemically with anti-lectin antibodies using the PAP method on Bouin and formalin-fixed paraffin sections of bovine abomasum during the fetal (14 to 27 wk) and postnatal period. In addition, gastrin and somatostatin were examined by the PAP method in serial sections of the same material. Dolichos biflorus agglutinin (DEA), which recognizes Nac-D-galactosamine residue, and Ulex europaeus I aglutinin (UEG I), which recognizes the L-fucose residue, yielded the most characteristic distribution patterns during the development of mucosa of bovine abomasum. DBAas marker was particularly involved in the process of differentiation of the fundic and pyloric part of the glands with the highest cytoplasmic concentration in enteroendocrine cells and in parietal cells and much less in the luminal surface, cell membranes and cytoplasm of other gland cells. The strong cytoplasmic appearance of DBA in endocrine cells coincided with the increased frequency of gastrin cells at 22 wk of gestation. UEA I was first uniforrnly localized in the surface mernbranes of the developing glands and later in cytoplasm of basal nest cells. A granular cytoplasmic staining gradually moved from the bottom towards the apical portion of glands with a predominant supranuclear localization accompanied by a strong luminal surface staining. Con A (D-glucose ; D-mannose), PNA (D-galactose) and WGA (Nac-D-glucosamine) receptors were mainly localized in the luminal surfaces and plasma membranes of developing glands with out marked affinity to the cytoplasm of gland cells during differentiation. The multistep process of development of mucosa in bovine abomasum is apparently accompanied by cell-to-cell interactions and cytoplasmic differentiation of gland cells determined by glycoconjugates and presumably by trophic action of gastrin.
