Morphometric measurements of Langerhans cells in irritant contact dermatitis
Langerhans cells (LCs) in the guinea pig skin can be demonstrated with histoenzymatic methods for both Mg++ and Ca++ activated adenosine triphosphatase (ATP-ase). The later was used for morphometric evaluation of the nurnber of LCs per square millimeter in the epidermis before and after irritation with 1% aqueous phenol solution. Our experiment for the first time demonstrated a stronger activity of CAE activated ATP-ASE in Langerhans cells, but the changes of LCs after irritation are more expressed in enzyme activity pattern of ATP-ase rather than in the frequency of Langerhans cells. The significant drop of the enzyme activity and loss of LCs were maximal at 24 hours after irritation, when the number of LCs per mm2 was 18.1±1. One month after treatment a linear, continual arrangement of LCs was re-established in the epidermis and the number of LCs per mm2 was 20.4± 2.
