Inhibition of Growth and Aflatoxin Accumulation of Aspergillus flavus by Using Newly Synthesized Analogues of Dehydroacetic Acid
The potential for inhibition of aflatoxin accumulation by aflatoxigenic fungus Aspergillus flavus ATCC 26949 was investigated using dehydroacetic acid (DHA) and the newly synthesized analogues, 4-hydroxy-3-(p-toluoyl)-6-(p-tolil)-2H-pyrane-2-one (DHT) and 5-Bromo-4-hydroxy-3-(p-toluoyl)-6-(p-tolil)-H-pyrane-2-one (BrDHT) on growth and aflatoxin B1 (AFB1) accumulation by mold Aspergillus flavus ATCC 26949 was studied in yeast-extract-sucrose (YES) medium to determine the possible use of these compounds as a means of controlling AFB1 accumulation.Experiments were carried out in a stationary culture at temperatures of 20 and 28 oC, respectively during 15 days.Mycelial dry weights were dcetermined gravimetrically, and concentration of AFB1 was quantitated by ELISA method.Concentrations of DHA and DHT of 0.1 and 0.2 mmol x L-1 were stimulated but concentrations higher than 0.5 mmol x L-1 of these compounds, mold growth and AFB1 synthesis partially inhibited.In experiments with 0.2 mmol x L-1 of BrDHT mold growth and synthesis of AFB1 were inhibited completely depending on the time and temperature of cultivation.
