Reproductive toxicity of atrazine given in vivo to female rats

The effect of 2-chloro-4-ethylamino-6-isopropylamino-s-triazine (atrazine, A) has been examined on the rat basal hypothalamus (H), pituitary (P), ovaries (O) and uterus (U). Immunohistichemical detection and quantitative evaluation of LH, FSH or estrogen receptor positive cells have shown significant effects of A in all examined tissues. Adult virgin females Wistar rats 100 days old were used in study. LH and FSH have been in P of control rats from 15-23% higher than in A treated. LH secreted from P was located at oocyte in O of control rats. Three days A treated rats (120 mg/kg b.w.) had regular estrus cycle and oocytes with a lot of LH, but seven days treated rats were in permanent diestrus and did not develop a mature oocyte. Estrogen receptor positive cells in U dependent upon of phase of cycle. After A treatment (our published results, Fd Chem Toxic, 1992) the number of binding sites in U cytosol for estradiol receptors have beeen reduced for 14-40.5%. Estrogen receptor positive cells in this study are reduced 23.5%. It probably leads to decrease production of mRNA and some proteins coded by genes in rat uterus.
