Parapoxvirus-induced modulation of porcine gut immune cells primed with Escherichia coli antigens

We attempted to determine whether or not the preparation of attenuated Parapoxvirus ovis, i. e. Baypamun (BPM) is capable of stimulating porcine lymphocytes from the jejunal lamina propria (JLP), ileal Peyer's patches (IPP), and the mesenteric lymph node (MLN) primed with the antigens of F4ac^+ enterotoxigenic Escherichia coli (ETEC). Five 4-week-old pigs were orally inoculated with 10^10 CFU of F4ac^+ ETEC strain M1823 in 60 ml of Trypticase soy broth (TSB). The control pigs were given the TSB only. At postinoculation Day 6 the pigs were killed, their gut lymphocytes were isolated, and treated in vitro with BPM, as well as with phytohemagglutinin (PHA), lipopolysaccharide (LPS), or peptidoglycan monomer (PGM), used as the control stimulators. Also, the patterns of cluster of differentiation (CD) antigen expression on T and B cells in the single cell suspensions from JLP, IPP, and MLN were determined by flow cytometry using anti-swine CD-specific monoclonal antibodies. BPM strongly stimulated the cells from JLP (P<0.01), IPP (P<0.05) and MLN (P<0.01) of pigs inoculated with strain M1823. Consistent with this is a better responsiveness of lymphocytes from IPP and MLN when PHA (P<0.01) or LPS (P<0.01) were applied. However, PGM failed to induce the lymphocyte proliferation in tested samples of lymphatic tissues. An increase of CD2a^+ and CD8a^+ T cells in JLP, and species-specific SWC1^+ T cells in MLN (P<0.05) was detected in M1823-treated pigs. In conclusion, BPM exhibited stimulatory properties of an immune response modifier, at least when applied in vitro, and thus, could be potentially synergistic when used in vivo in the vaccination programs to control the postweaning colibacillosis in swine.
