Distribution of cytolytic and suppressor T cell subsets in the small intestine of pigs demonstrated by immunohistochemical methods
The aim of study was to demonstrate the phenotypic diversity of jejunal/ileal cytolytic and suppressor T cell subsets in 8-week-old pigs by using either avidine-biotin complex (ABC) or peroxidase-antiperoxidase (PAP) immunohistochemical methods on tissues. Tissue samples of the small intestine were fixed in methanol-chloroform-acetic acid and embedded in paraplast. With monoclonal antibody specific for CD3a membrane antigen used in ABC method, we have demonstrated the presence of that epitope in the jejunum and ileum of investigated animals . Using polyclonal antibodies specific for S-100 protein in PAP method we have demonstrated also the presents of S-100 positive cells in the jejunum and ileum of the same pigs. In the jejunum we have demonstrated that the most frequent jejunal CD3a^+ cells were intraepithelial lymphocytes. In the ileum we have found numerous clusters of CD3a^+T lymphocytes between Peyers patches follicles and solitary CD3a^+ cells in the follicles. The most frequent S-100^+ T cells were found in jejunal/ileal lamina propria, particularly between the crypts. If S-100 protein binds to porcine CD3a^+CD8^+CD4^ suppressor T cells, then it would be possible by identifying CD3a^+ and S-100^+ cells in paraplast embedded tissue sections, to established the distribution cytolytic CD3a^+CD8^+CD4^S-100^ and suppressor CD3a^+CD8^+CD4^S-100^+ T lymphocytes in the intestinal mucosa of pigs. This would implicate the patterns and possible results of local immune reaction to dietary antigens or intestinal pathogens.
