In vivo immunogenic tetanus toxoid correlate of in vitro induced equine lymphocyte proliferation

Equine antibody against tetanus is still widely used in nondeveloped countries since human antitoxin immunoglobulins have not proved any advantage in the therapy of neonates diseased by tetanus. There are suggestive evidences that in vitro test using tetanus toxoid (TTd) and common mitogens (PHA and PWM) could be of the practical value in classifying the horses intended for antitetanus serum production as low or high responders. The aim of current study was to (1) evaluate this hypothesis, and (2) determine the immunogenicity of BaypamunŇ (BPM) in the immunization of horses with TTd and tetanus toxin (TT). Ten horses were intramuscularly immunized with either TTd and TT or TTd/TT combined with two doses of 2 ml of BPM given subcutaneously. The animals were bled before the immunization and after each immunization step, their PBL were isolated and stimulated with either 10 (mikro)l/ml of PHA or PWM, or 20 (micro)l/ml of BPM, and the reactivity of PBL was expressed as the stimulation index. TTd/TT-treated horses had 2 times lower PBL responses to PWM at the end of the immunization than they have had as nontreated animals before the immunization. Conversely, PBL from TTd-treated BPM-stimulated horses exhibited about 2 times higher reactivity to PWM. The hyperimmunization with TT did not affect PBL responsiveness to PWM. Also, the titers of specific antibody were significantly increased (P < 0.05) in the horses given BPM along with TTd. Considering these results, as well as our previous data, it could be possible to select horses, the donors of antitetanus serum according to their PBL reactivity to PWM. Their productivity in plasmapheresis could be additionally improved by including BPM in the immunization schedule.
