PSMAD 1/5/8 is expressed in normal human kidney as well as in renal cell carcinoma
INTRODUCTION Bone morphogenetic proteins are multifuncional growth factors involved in development, morphogenesis, cell proliferation and apoptosis. Signaling mediated by the BMPs transduce proteins called Smads after activation of serine-threonine kinase receptor, type I receptor and type II receptor. BMPs belong to TGF-β family which is known to promote tumorogenesis. Also BMPs and their signaling pathway have been investigated in different tumor types and it has been shown that levels of expression correlate with the degree of malignancy. Interestingly, in some cases they act as a tumor suppressor that promotes apoptosis. Signaling BMP pathway conducted through Smad 1/5/8 in renal carcinoma has not been investigated yet. AIM The objective of this study was to determine whether and where BMP signaling is disrupted in renal carcinoma. MATERIALS AND METHODS All tissue specimens were obtained after nephrectomy and fixed in formalin. Parafin embeded sections were immunohistochemicaly stained with pSmad 1/5/8 antibody (Cell Signaling). We performed antigen retrieval in 0.1M citric acid, pH 7.2, for 15 minutes in microwave, antibody was applied in 1:100 dilution overnight at 4°C. RESULTS The expression of pSmad 1/5/8 was investigated in tissue samples of morphologicaly normal kidney tissue taken from distant site in nephrectomized kidney, tissue adjacent to carcinoma and renal carcinoma. In normal tissue and tissue that surrounds carcinoma the expression of pSmad was localized in the cytoplasm and nuclei of tubular cells. pSmad was expressed in all normal and adjacent to carcinoma tissue samples. We also found the activity of BMP signaling pathway in renal carcinoma cells what was expressed through positive staining for pSmad 1/5/8. Immunostaining was localized in citoplasm and nucleus, too. CONCLUSION Theese results indicate that BMP signaling pathway is active in renal carcinoma as well as in tubular cells of normal kidney.
