Genetic Relations Among Basil Taxa (Ocimum L.) Based on Molecular Markers, Nuclear DNA Content and Chromosome Number

The phylogeny and taxonomy of genus Ocimum is somewhat confusing due to hybridization, polyploidy and the existence of chemotypes or chemical races that do not differ significantly in morphology. Twenty eight basil accessions including six Ocimum species and six botanical varieties and cultivars of O. basilicum were studied using molecular markers, nuclear DNA content analysis and chromosome counting. The results of nuclear DNA content and chromosome numbers supported grouping of Ocimum species revealed by molecular markers. The results indicate that two O. basilicum var. purpurascens accessions, O. x citriodorum and O. americanum could represent allopolyploid species that share some common parental genomes. Ocimum tenuiflorum, although being clustered together with O. gratissimum on both phenetic trees was according to genetic distance and nuclear DNA content most distant form other studied accessions. In this paper we have demonstrated that RAPD and AFLP analyses combined with chromosome number and nuclear DNA content information could contribute in resolving existing problems in the phylogeny and taxonomy of basil taxa.
