Ikaros family transcription factors in chronic and acute leukemia
Ikaros transcription factors, Ikaros, Aiolos and Helios, are important regulators of lymphocyte differentiation and the recognition of their expression pattern might help to understand the mechanisms of leukemia development. In order to determine whether Ikaros family members have a role in human leukemia we analyzed primary leukemic cells for differential expression of Ikaros, Aiolos and Helios mRNA by means of real time quantitative RT-PCR method. We found that the relative quantity of Ikaros and Helios mRNA was similar in all subgroups. Though, analysis of Aiolos, showed a clear difference among groups due to its lower expression in all types of acute leukemia investigated. Lower Aiolos expression seems to be related to the cell developmental stage and this would give an explanation of its decreased expression in acute leukemia as compared to the expression in normal peripheral blood cells and/or in chronic leukemia cells (CLL). Our results indicate that Aiolos mRNA level might represent a cell maturational stage and in this context it potentially could be useful as a target for stimulating cell differentiation.
