Effects of cigarette smoke extract on induction of TIMP-1, MMP-9 and MMP-2 in human A549 cell line
Chronic obstructive pulmonary disease (COPD) is the most frequent chronic respiratory disease with the progressive course. It is characterised with the development of local and systemic inflammation. Cigarette smoking is the main etiologic factor causing the inflammatory response in COPD patients. Matrix metalloproteinase -9 and -2 (MMP-9 and MMP-2) can play an important role in inflammation and lung tissue remodelling owing to their gelatinolitic activity. Tissue inhibitor of metalloproteinases-1 (TIMP-1) is an inducible protein and it is a potent inhibitor of matrix metalloproteinases, including MMP-9 and MMP-2. An improper balance of MMP and TIMP production correlates with different pathological conditions. In this study the effect of cigarette smoke extract (CSE) on induction of TIMP-1, MMP-9 and MMP-2 in human alveolar epithelial cell line (A549) was examined. Human A549 cells were cultured in F-12 culture medium and exposed to different concentrations of CSE (1.25, 2.5, 5, 10 and 20%) for 4h and 24h, respectively. Quantitative determination of secreted TIMP-1, MMP-9 and MMP-2 was performed in supernatant of culture media, using the ELISA kit. The results show very low secretion of MMP-9 and MMP-2 in A549 cells after 4h incubation with CSE. However, CSE induces significant release of MMP-2 from A549 cells after 24h incubation (increase for 114.1 % when compared with control sample, p<0.05). At the same time, release of TIMP-1 was also stimulated (increase for 13.5 % when compared with control sample, p<0.05) after 24h incubation. In contrast to our expectations, there was no induction of MMP-9 with any concentration of CSE. Results are showing that CSE is stimulating release of MMP-2 from A549 cells, but concomitant increase of TIMP-1 release might indicate activation of the defence mechanisms against CSE produced tissue damage.
