Comparison between ELISA and TLC methods for determination of fumonisins (B1+B2+B3) and aflatoxins (B1+B2+G1+G2) in food and feed

Fusarium and Aspergillus species frequently contaminate crops in the field and storage. Therefore, fumonisins (FBs) and aflatoxins (AFs) could be found in these samples in wide range of concentrations depending on environmental and storage conditions. Consumption of FB1-contaminated feed has been associated with various diseases in animals including leukoencephalomalacia in horses, porcine pulmonary oedema, immunosuppression, liver and kidney toxicity and liver cancer and was connected with human oesophageal carcinoma in Southern Africa and China. Aflatoxins are well-known hepatotoxins and carcinogens for animals and humans. The aim of this study was to determine the frequency and concentrations of FBs (B1+B2+B3) and AFs (B1+B2+G1+G2) in cereal and feed samples (N=37) randomly collected in individual households of endemic nephropathy region (EN) in Croatia (2007). Direct competitive ELISA test kit (Veratox ) was used for FBs and AFs determination according to manufacturer instructions (Neogen Europe Ltd). FBs were detected in 27% of samples in range between 200 and 20700  g/kg (mean 3690  g/kg), while AFs were recovered from 40.5% of samples at concentrations ranging from 1.1  g/kg to 10.3  g/kg (mean 3.28  g/kg). FBs and AFs were simultaneously occurred in 13.5 % of samples. These mycotoxins in cereals and feed were under the permissible levels according to Croatian law (NN/118/2007) and European Commission (EC/576/2006 and EC/1881/2006) except one sample of maize attended for human consumption which exceeded tolerable level of FBs. Concentrations of FBs and AFs detected by ELISA significantly correlate with results obtained by TLC. ELISA provides an attractive and promising method for FBs and AFs detection in food due to high specificity, sensitivity, simplicity and analysis of high number of samples.
