It is well known that sea-surface microlayers play an important role in the marine environmental protection and global change Sea surface layer is generally enriched in organic substances, particularly those which are surface active, metal ions, bacteria and other microorganisms, relative to subsurface water. Although the physical and chemical properties of sea-surface microlayers have been studied extensively, there is still a lack of knowledge about physico-chemical processes governing the formation and properties of the surface microlayers. Sea-surface microlayers can be studied either in original sample without any pretreatment or they can been previously extracted by organic solvent and then investigated as ex-situ reconstructed film. Physico-chemical characteristics of natural and ex-situ reconstructed sea surface microlayer samples were studied using a complex methodological approach. Surface microlayer samples (both sea and lake) and the underlying water were collected in different seasons and different weather conditions in the northern most part of the Adriatic sea, as well as in the region of Middle Adriatic. Here we present basic principles of the methods and illustrate their application on characterization of a natural sea surface microlayer sample.
