Structures and functional roles of transcription regulators in the circadian system of Bombyx mori
In the presented study it was investigated the diversity in the molecular mechanisms of circadian rhythms in the silkworm, Bombyx mori to provide deeper insights into the mechanisms of the clock genes function in silkmoths. It is specially focused on molecular cloning and characterization of the transcription factor regulator CYCLE in the silkworm, B. mori its structure and expression pattern by using several molecular methods (Polymerase chain reaction-PCR, screening a cDNA library (colony hybridization), RT-PCR, and Northern blot analysis). In addition, it examined temporal expression patterns of mRNA abundance of B. mori Clock and cycle genes in the head of adult moths by Northern blot analysis. This work makes also immunohistochemical investigation on distributions of the main circadian clock-related proteins in the cephalic nervous system of B. mori. Moreover, this examination included around-the-clock semi-quantitative immunocytochemical analysis on expression of these proteins, and double-labeling experiments for co-localization of the proteins in the cephalic ganglion of the silkmoth, B. mori.
