Mjerenje apsorbirane doze zračenja i oštećenja DNK pri RTG pregledu pluća kod djece

The knowledge of absorbed dose in tissues is important in radiological department especially in paediatrics to estimate the risk of radiation exposure. In addition the risk of genome damage can also be estimated by using several cytogenetic techniques. In this paper comet assay was chosen as suitable technique for assessment of DNA damage induced by ionising radiation of chest radiogram. The aim of this study was to correlate the data on the cytogenetic status of children caused by X-ray examinations of chest with the exposure dose. To compare these data to the dose of exposure determined with thermoluminescence (TL) dosimetry and the radiophotoluminescent (RPL) glass dosimetry system, teen children with pulmonary diseases, ages between 4 and 14 years were assessed. Samples of peripheral blood were collected from patients before and after X-ray examination and Comet Assay were carried out under alkaline conditions using tail length as a measure of DNA damage. The differences between the mean values of tail length for Comet assay before (13.71± ; 0.60) and after (14.51± ; 0.81) exposure were statistically significant (P<0.05, ANOVA). The correlation between doses in the primary beam (measured on the back of children patients) and the ratio of tail lengths (DNA damage) after and before irradiation was found. The doses measured with TL and RPL dosimeters show an agreement satisfactory for X-ray diagnostics and are suitable for dosimetric measurements in X-ray diagnostic.
