The effects of organophosphorous compounds on precursors of human skeletal muscle regeneration

Besides well established effects of organophosphorous (OPs) compounds exerted through inhibition of cholinesterases (ChE), ChE are not the only targets of OPs and various intracellular mechanisms are modified due to direct or indirect OPs actions. Typically met upon the OPs poisoning are myopathies, which might be a consequence of insufficient muscle regeneration. It is therefore important to know whether OPs in any way interfere with the mechanisms involved in muscle regeneration process. The proposed mechanisms of OPs effects on muscle regeneration process include conformational changes of ChE protein which might affect so called noncholinergic acetylcholinesterase (AChE) functions or direct organophosphate binding to some of non-cholinergic molecules. The most important step in the process of muscle regeneration is myoblast proliferation, myoblast fusion and myotube innervation. Our investigations on the effects of OPs on the muscle regeneration were therefore focused on these early stages of muscle regeneration. We studied the effects of OPs on the expression of AChE, heat-shock protein (HSP) 70 and 27, interleukin-6, known for their roles in stress response of the skeletal muscle, the effects on the expression of Na+/K+ATPase protein subunits, which are markers of muscle differentiation. We found that exposure of muscle cells to OPs did not interfere with the processes of myoblast fusion and innervation and AChE expression. In cells exposed to OPs constitutive interleukin-6 secretion was inhibited. The levels of Na+/K+ATPase α 2 subunit and HSP 70 and 27 were higher than in controls. These effects might contribute to the myopatic effects known to accompany OPs poisoning.
