FLUORESCENCE – PRINCIPLES AND APPLICATIONS
This lecture will give an introduction to the luminescence methods of molecular spectroscopy: fluorescence, phosphorescence, and chemiluminescence [1, 2]. The common principle of these techniques is the measurement of the emission spectra obtained when excited molecules decay to their ground states. Compared to absorption techniques, luminescence methods are extremely sensitive, with detection limits often one to three orders of magnitude smaller than those encountered in absorption spectroscopy. However, the number of substances which emitted light is limited. The brief overview of the fluorescence mechanism and quantitative fluorimetry (fluorescence efficiency or quantum yield, the inner filter effect, and quenching) will be given. The analytical methods for organic and inorganic analytes determination, such as direct, derivatisation and quenching methods will be discussed as well as the application of fluorescence in liquid chromatography and immunoassays.
