Biotransformation of 5alpha- and 5beta-androstane-3beta/3alpha, 17beta- -diols by human CYP3A4 and 2C19 enzymes
Cytochrome P450 (CYP) enzymes catalyze biotransformations of endogenous and exogenous steroids in regio- and stereoselective manner both in vitro and in vivo. Of the identified human CYP enzymes, the four subfamilies (CYP1A, CYP2B, CYP2C and CYP3A) catalyze hydroxylation of the steroids selectively at different positions (1-4). In the present study, 5?-androstane-3?/?,17?-diols, and 5?-androstane-3?,17?-diol were incubated with human recombinant CYP3A4 and CYP2C19 enzymes (PANVERA, USA). Structures of metabolites, as trimethylsilyl-ether derivatives, were determined by the GC/MS method, and the major metabolites were quantified (3,4). 5?-Dihydrotestosterone (5?-DHT) was formed selectively from 5?-androstane-3?,17?-diol by both CYP3A4 and CYP2C9 enzymes, higher level of formation was observed with the former enzyme.
