RATIONALE: Several studies reported spontaneous circling behavior in inbred rat strains, which was attributed to the dopaminergic dysfunction (Loscher et al., Neuroscience 74: 1996 ; 1135-42, Lessenich et al. Neuroscience107: 2001 ; 615-28). Here we present novel phenotype of unilateral spontaneous circling behavior found among inbred Wistar rats. OBJECTIVES: to describe behavioral pattern of affected rats, determine possible line of inheritance and to investigate behavioral response to drug administration. MATERIALS AND METHODS: Open filed, tail-hanging and swimming test were used to asses motor abnormalities before and after administration of following agents: bromocriptine (10 mg/kg, i.p.) ; trihexyphenidyl (1.5 mg/kg, i.p.) ; clozapine (10 mg/kg, i.p.) ; haloperidol (5 mg/kg, i.p.) and sulpirid (100 mg/kg, i.p.) in affected and matched control rats. For the mating experiment circling-circling and circling-normal rats were paired. RESULTS: Phenotype: Affected rats of both sex exhibit intensive unilateral circling with strong and permanent head distortion towards the side of rotation. Circling occurs spontaneously or as a response to stress. Symptoms emerge between 2.5 and 5.5 months of age. Inheritance: Only 2 out of 38 offspring developed phenotype behavior. Behavioral tests and pharmacology: Affected rats weren’ t able to swim. In open field rats exhibited normal locomotor activity interrupted with phases of circling. In tail-hanging test rats exhibited extensive rotations. Landing response was damaged. Only haloperidol and sulpirid had significant effect to behaviors observed: circling was completely abolished, landing response was normal, 85% of affected rats were able to swim. CONCLUSION: Phenotype of Zagreb spontaneously unilateral circling rats differs from ones previously reported. Line of inheritance remains unknown. First pharmacological tests indicate that circling may be associated with dopaminergic D2 receptors.
