THE INVOLVEMENT OF PERFORIN EXPRESSION IN PERIPHERAL BLOOD OF BURNED PATIENTS
Introduction: Cytotoxic T cells (CTL) and natural killer (NK) cells induce cell death of their target cells either by the release of perforin and granzymes or by the surface interaction between Fas ligand and Fas. Both these lytic pathways induce apoptosis of neoplastic and an altered-self cell. Methods: Venous blood samples were taken and peripheral blood leukocytes were isolated. Phenotype of lymphocytes and perforin expression were analyzed using intracellular and surface immunofluoresecncy and flow cytometry. Results: Our data demonstrated that 10-15% of PBL in patients are perforin positive. Percentage of all double positive cells showed significant decrease in patients with burns injuries in comparation with healthy volunteers, showing the possible prognostic recovery value of these findings. Conclusions: Our results suggest the important role of perforin positive cells in naï ; ve immune response and cytotoxic activity of peripheral blood cells of patients with burn injuries.
