Nitric oxide synthase in the hypothalamus of alloxan-diabetic rats.
Alterations in neuronal nitric oxide synthase (nNOS) and nitric oxide (NO) mediated transmission in diabetic animals have already been described in the peripherla nervous system, but no data have been reported fot the central nervous system. We have investigated the expression of n NOS mRNA in paraventricular (PARVN), periventricular (PERVN) and supraoptic (SON) hypothalamic nuclei of alloxan-diabetic rats using in situ hybridization with 35D labelled synthetic oligonucleotides. The expression of nNOS mRNA was greatly increased in the magnocellular part of PARVN and in SON of 1 week (+ 118% and +79%, p<0.05) and 4 week (+92% and +78%, py0.05) alloxan-diabetic animals, respectively. Our results suggest that elevated hypothalamic NO production and neurotoxicity may underlie brain pathologies observed in diabetes mellitus.
