Expression of bacteriophage derived scFv and their incorporation into third generation porphyrin immunoconjugates for targeted PDT

An expression system giving high yield of scFv fragments has been established. The molar ratios at which the conjugation to scFv was carried out has been optimized. These immunoconjugates showed low levels of non-covalently associated porphyrin, minimizing potential non-specific cytotoxicity. Based on these findings, together with preliminary data using immunoconjugates of scFv raised against freshly-isolated tumour cells, we suggest that scFv:poprhyrin conjugates may show promise for use as an antibody-targeted photodynamic strategy.
