Effect of differnt amine fluoride concentrations on enamel surface studied by scanning electron miicroscopy

The mechanisms of the action of fluoride in caries prevention are only partially understood. The fluoride can be incorporated in enamel in a soluble (CaF2) and/or structurally bound (fluoroapatite) form. Calcium fluoride is the major product deposited on enamel after exposure to high concentrations of topically applied fluoride. CaF2 has been visualized by scanning electron microscope (SEM) as spherical globules on enamel. This in vitro study was designed to compare the enamel surface morphology after topical applications of the amine fluoride solutions with different fluoride content (1.0, 0.5, and 0.25% F). The evaluation of the interaction of the amine fluoride solutions with sound enamel was done with a new SEM/EDX technique. Enamel slabs were cut from buccal surfaces of extracted sound third molars, randomly distributed into four groups corresponding to the treatment regimens. The SEM technique showed the globular, crystalline and amorphous material on the enamel surfaces after the 3-min. application of the different fluoride concentrations per three days. A difference was seen between the enamel surfaces treated with the amine fluoride solutions and the control group. However, the impression was that the distribution of the deposits was more homogeneous in groups treated with higher fluoride concentrations. The EDX analysis showed no differences in relative intensity of the Ca/F ratio after 0.5 and 1 % F treatment. The conclusion is that interaction of the 1.0 and 0.5% topical fluoride concentrations and sound enamel surfaces can induce the same alternation of surface morphology and a similar degree of protection.
