Dynamics of body temperature changes in rats induced by chlorpromazine and clozapine
Many drugs influence temperature balance, especially those acting on the central nervous system. Therefore, in this work a detailed characterization of dynamics of hypothermic changes induced by chlorpromazine and clozapine was investigated on anesthetized rats. The results showed that both drugs induced temperature decreases, the duration and intensity of which were dose-dependent. Even the lowest doses of chlorpromazine (0.7 mg/kg b.m., n=7) and clozapine (4.2 mg/kg b.m., n=7) applied i.p. caused hypothermia that lasted about five hours. The maximal temperature differences induced by these doses of chlorpromazine and clozapine were 2.5 +/-0.4 C, respectively. Chlorpromazine induced higher temperature decreases than clozapine in all other applied doses. These results suggest that in practice special precautions are required, particularly for the long-term application of higher doses of these drugs in elderly patients who might have multiple complications because of hypothermia.
